Holy crap, I totally forgot about Sluggy Freelance! Wow, that goes back a couple of years.

I read Penny Arcade daily - it's not for all, but it works for me.

The greatest webcomic ever is Order of the Stick.  You have to be a D&D geek to understand it, but it's funny as hell.

Hells no, the greatest ever for me is Penny Arcade. Their humor works for me, and I think the two guys are funny as all hell.

My current cue besides the ones already mentioned.

    Applegeeks
    Mac Hall
    Little Gamers
    Theater Hopper
Instant Classic Currently on Hiatus due to health problems
Life of Riley It's dead but it still deserves mention. Especially the Bob Wars.
Errant Story and Exploitation Now before that.
    VG Cats
    Kill Harry
Force Monkeys Another archive but still excellent.
NSFWSexy Losers There's a lot of different strips that follow the same theme if you will. Again it is NSFW[/list]

    Burninate!

Nobody ever gets the joke when I use that word in real life, though some do find the word itself amusing.

    Achewood for the win.

You do need to read some back issues to 'get it,' though.

I would have to second two of the above.

Something Positive is a great strip that has been consistently hilarious for 3 years now.  The current storyline involves three friend's quests to recover a childhood toy that has been converted into a furry plaything.  Yes, *that* kind of furry.  This is one that I would suggest starting from day 1 just so you get to know the characters as the author introduced them.  The comic is fantastically mysanthropic.  If you think Garfield is too bitter to be funny and that Peanuts is quality, this is not the comic for you.

Sexy Losers is another great one but it is NSFW.  Let me repeat that.  NSFW.  NSFW.  NSFW.  It deals with masturbation, incest, necrophilia, a dildo and a reanimated corpse that is tired of being used as a sex toy.  Perhaps you are getting a clue why you shouldn't check this one out at work or anywhere kids might be nearby.  That said, it is very well drawn and absolutely hilarious.  I went through its archives in one night and didn't stop laughing the whole time.  If you think you can handle this, you owe it to yourself to check it out.
