Jude Law as the mentally challenged confederate soldier in Cold Mountain. He showed a lot of depth to both master a north carolinian accent and to convincingly act retarded.
Oh wait, you said best, my bad.

Everybody in Lord of the Rings was excellently casted, which is one prime reason the movies are so great. Only the Eowyn character would I consider somebody else for the role.
Edward Norton in American history X. Tom Hanks in Forest Gump Arnold as Conan the barbarian

I agree with alot of the earlier ones posted, i.e. Ian Mckellar, Denzel as Malcom etc. Here are a few I did not see posted.
Kurt Russell as Jack Burton - Big Trouble in Little China Victor Wong as Egg Shen - Big Trouble in Little China John Wayne - about 90% of all his films but .... Rooster Cogburn in Rooster Cogburn Jimmy Stewart - again just about anything however, Elwood P Dowd in Harvey Carey Grant - see above, David Huxley in Bringing up Baby, Mortimer Brewster in Arsenic and Old Lace Danny Kaye as Hubert Hawkins in the Court Jester Hugo Weaving as Agent Smith in Matrix 1 & 3 Stanley Tucci as "Muerte" in Undercover Blues(this movie sucked other then Tucci). Jeff Bridges as "The Dude" in Big Lebowski John Goodman as Walter Sobchak in Big Lewbowski Nicholas Cage as H.I. Mcdunnogh in Raising Arizona Dustin Hoffman as Raymond Babbitt in Rain Man Michael Keaton as Beetlejuice in Beetlejuice Antonio Banderas as El Mariachi in Desperado John Travolta as Chili Palmer in Get Shorty Jason Mewes as "Jay" in any of Kevin Smiths movies
I listed actresses on a seperate list, I'll post later.

Greatest casting jobs of all time:
3) Michael keaton as Beetlejuice (Damn someone beat me to it!! Greatest Oscar Snub ever. I'm serious) 2) Rutger Hauer as Roy Batty. I want more life, fucker! 1) Christopher Lee as Saruman. Good..God...He looks like an old wizard, with or without robes.

Honorable Mentions: Val Kilmer as sick and dying Doc Holliday Ed Harris:90% of his career, but best as the Hostile saleman in Glengaryglenross...speaking of which: old Jack Lemmon as Shelley the Machine Levine, GGGR. Alec Baldwin (cameo) in GGGR. John Cusack:90% of his career, possibly best as Lane Meyer in Better Off Dead (reallly, can you see anyone else doing that role?) Venron Wells as The Might Wez (Road Warrior) Arnold as Conan (Automatic) The original casting of the Terminator: Michael Biehn as The Terminator, Arnold as the future commando sent back to stop him. Lance Henrikson as Bishop making his debut as a colossal fatass - Marlon Brando as Kurtz

I'm serious) Another all-time great oscar snub, and best casting entry would be Val Kilmer as Jim Morrison in The Doors. Kilmer looked just liked him, acted just like him, and even sang the songs himself, and it's hard to tell the difference there, too. It was a brilliant performance.
